HPLC Testing Explained: How Peptide Purity Is Actually Measured

Every serious purity claim in the research peptide industry ultimately traces back to one testing method: HPLC, or High-Performance Liquid Chromatography. Understanding roughly how it works makes it much easier to evaluate whether a supplier's purity claim is actually meaningful.
What HPLC Actually Does
HPLC works by pushing a liquid sample through a column packed with a material that interacts differently with different molecules. As the sample passes through, individual components separate based on their chemical properties and exit the column at different times. A detector records each component as it exits, producing a chromatogram — a graph showing distinct peaks for each substance present in the sample.
How This Translates to a Purity Percentage
In a peptide purity test, the goal is to see one dominant peak corresponding to the intended compound. The area under that peak, relative to the total area of all peaks detected, gives the purity percentage. A 99% purity result means the target peptide's peak accounts for 99% of the total detected material, with the remaining 1% representing synthesis byproducts or other impurities.
HPLC offers a level of resolution that allows labs to detect even small amounts of related impurities — compounds structurally similar to the target peptide that might otherwise go unnoticed with less precise testing methods.
What HPLC Doesn't Tell You
HPLC purity results don't automatically capture net peptide content, which accounts for non-peptide mass like moisture and counter-ions. A complete testing package often pairs HPLC with mass spectrometry, which confirms molecular identity, giving a more complete picture than purity percentage alone.

- Was the purity figure generated via HPLC specifically?
- Is the chromatogram itself available, or just the summary percentage?
- Was testing performed by an independent lab, or in-house?
Why This Matters Beyond Curiosity
Understanding HPLC helps researchers ask sharper, more specific questions when evaluating a Certificate of Analysis, rather than accepting a purity percentage at face value without understanding how it was generated.
